Both Specific Endothelial and Proximal Tubular Adam17 Deletion Protect against Diabetic Nephropathy

چکیده
ADAM17 is a disintegrin and metalloproteinase capable of cleaving the ectodomains diverse variety molecules including TNF-?, TGF-?, L-selectin, ACE2. We have previously demonstrated that renal upregulated in diabetic mice. The role endothelial (eAdam17) proximal tubular (tAdam17) Adam17 deletion histology, modulation renin angiotensin system (RAS), inflammation, fibrosis was studied mouse model type 1 Diabetes Mellitus. Moreover, effect an vitro 3D cell culture from human cells under high glucose conditions evaluated. eAdam17 attenuates whereas tAdam17 decreases podocyte loss, RAS, macrophage infiltration, ?-SMA collagen accumulation. reinforced findings obtained tAdam17KO mice with decreased knockout spheroids. In conclusion, either or mitigates kidney injury by targeting different pathways. manipulation should be considered as therapeutic strategy for treating DN.

Inhibition of Kidney Proximal Tubular Glucose Reabsorption Does Not Prevent against Diabetic Nephropathy in Type 1 Diabetic eNOS Knockout Mice
BACKGROUND AND OBJECTIVE Sodium glucose cotransporter 2 (SGLT2) is the main luminal glucose transporter in the kidney. SGLT2 inhibition results in glycosuria and improved glycaemic control. Drugs inhibiting this transporter have recently been approved for clinical use and have been suggested to have potential renoprotective benefits by limiting glycotoxicity in the proximal tubule. We aimed to ...

The rising incidence and prevalence of chronic kidney disease (CKD) (glomerular filtration rate ,60 mL/min per 1.73 m) is a major public health concern (U.S. Renal Data System 2010, http://www.usrds.org). Hypertension is the second leading cause of end-stage renal disease (ESRD) in the U.S. and is also a major contributor to diabetic renal disease, which is the leading cause of CKD and ESRD (U....
متن کاملEffects of SGLT2 Inhibition in Human Kidney Proximal Tubular Cells—Renoprotection in Diabetic Nephropathy?
Sodium/glucose cotransporter 2 (SGLT2) inhibitors are oral hypoglycemic agents used to treat patients with diabetes mellitus. SGLT2 inhibitors block reabsorption of filtered glucose by inhibiting SGLT2, the primary glucose transporter in the proximal tubular cell (PTC), leading to glycosuria and lowering of serum glucose. We examined the renoprotective effects of the SGLT2 inhibitor empaglifloz...
متن کاملThe pathogenic role of the renal proximal tubular cell in diabetic nephropathy.
A growing body of evidence indicates that the renal proximal tubular epithelial cell (PTEC) plays an important role in the pathogenesis of diabetic nephropathy (DN). Microalbuminuria that intensifies over time to overt proteinuria, a hallmark of DN, is already known to activate the PTEC to induce tubulointerstitial inflammation. In addition to proteins, a number of diabetic substrates including...
متن کاملEndothelial cilia protect against atherosclerosis.
Research over the past 15 years has established that defective primary cilia play a fundamental role in the pathogenesis of various genetic and acquired human diseases now termed ciliopathies. A new study, published in this issue of EMBO reports, expands the spectrum of ciliarelated diseases by uncovering a role for primary cilia in preventing cardiovascular disease. Dinsmore and Reiter show th...
